2-Mesitylenesulfonyl Chloride (2,4,6-Trimethylbenzenesulfonyl Chloride) (CAS: 773-64-8) is used as reagent for intramolecular lactamizations and lactonization. It also acts as a reagent for protecting guanidino groups and tryptophan. It is used as coupling reagent in polynucleotide synthesis.

Production method 30g(0.25mol) mesitylene was added dropwise to 80g(0.69mol) of chlorosulfonic acid cooled to 0℃. Over a period of 0.5h, and stirring was continued for a further period of h. The reaction mixture was poured into crushed ice to give 50g of 2,4,6-Trimethylbenzenesulfonyl Chloride in 92% yield.
