This product is 1-(3-Methoxypropyl)-4-Piperidinamine. Containing C9H20N2O shall not Less than 98.0%, calculated on the anhydrous basis.
[Properties: This product is colorless to light yellow liquid.
Take an appropriate amount of water of this product, add solvent (methanol), dissolve it, determine it according to the water determination method (Appendix Ⅷ M, first method A), use Fisher's test drops to determine until the solution changes from light yellow to red brown indicating the end point, and conduct another blank test. Calculate according to the following formula: the water content should not exceed 1.0%.
Moisture content (%) = (A-B) F/W×100%
Where, A is the volume of Fisher's liquid consumed by the test product (ml);
B is the volume of Fisher's test liquid consumed by blank space, ml;
F is the weight of Fisher's test solution equivalent to water per 1ml, mg;
W is the weight of the test product, mg
Single impurity
According to the detection conditions and methods under [content determination], the chromatogram was recorded and the retention time from the main peak was 3 times. If there is an impurity peak in the test solution, the single impurity shall be no more than 0.5% and the sum of all impurities shall be no more than 2.0% according to the area normalization method
[Content Determination] According to residual solvent determination method (Appendix Ⅷ P)
(6%) cyanopropyl phenyl - (94%) dimethylpolysiloxane (or similar polarity) as the fixed liquid capillary column;
The programmed warming method of capillary column headspace sampling system with gas chromatographic conditions was used. The initial temperature was 100℃ for 5min, and the temperature was raised at 15℃ per minute to 250℃ for 4min. The inlet temperature was 250℃, and the detector temperature was 280℃. The flow rate was 4ml/min. The shunt ratio is 8:1.
About 0.6g of this product was taken, accurately weighed, placed in a 10ml bottle, dissolved with methanol and diluted to the scale, as the test product solution. The sample solution was injected with 1μl for at least two consecutive times, respectively. The peak area of the peak to be measured was determined. The main peak purity was not less than 98.0% according to the peak area normalization method.

1-(3-Methoxypropyl)-4-Piperidinamine (CAS: 179474-79-4) is a useful synthetic intermediate in the synthesis of Prucalopride Succinate (CAS: 179474-85-2). Prucalopride is a selective 5-HT4 receptor agonist used effective for chronic constipation, but is not currently approved in the U.S. Prucalopride is approved for the treatment of chronic constipation in Europe. Prucalopride succinate, a first-in-class dihydrobenzofurancarboxamide, is a selective serotonin (5- HT4) receptor agonist. The drug, marketed under the brand name Resolor®, possesses enterokinetic activity and was developed by the Belgian-based pharmaceutical firm Movetis. Prucalopride alters colonic motility patterns via serotonin 5-HT4 receptor stimulation, triggering the central propulsive force for defecation.
