D-Prolinol (CAS: 68832-13-3), the D-enantiomer of prolinol, is a chiral amino alcohol used as a chiral resolver in organic synthesis. In recent years, with the rapid development of asymmetric catalysis technology, D-Prolinol has become an excellent ligand for metal coordination catalysis, widely used in asymmetric metal alkylation reaction, Michael addition reaction, phase transfer catalysis and other important organic synthesis. D-Prolinol makes use of an intermediate of pharmaceuticals.
Synthesis methods: There are two main methods for the synthesis of D-Prolinol: (1) Indirect reduction method: taking D-Proline as raw material, esterification, neutralization, and then reduction with NaBH4; (2) Direct reduction method: D-Proline was directly reduced by AlLiH4. The former process is complicated and the yield is low (42.6%). The latter operation is harsh, low temperature ice bath drops, high cost, deep product color. The author directly reduced D-proline to D-prolinol by using the reduction system of NaBH4 additive I2 (see the figure below), which has low cost, good quality, stable process, simple operation and easy industrialization.
