L-Glutamic Acid 5-tert-Butyl Ester (H-Glu(OtBu)-OH) (CAS: 2419-56-9) is a glutamic acid derivative, can be obtained by the reaction of glutamic acid and tert-butyl acetate.
Amino acids and derivatives, used in peptide synthesis, organic synthesis intermediates, pharmaceutical intermediates, biochemical reagents, chemical reagents.
Preparation add 2324g of tert-butyl acetate and 73.5g of glutamic acid into a 3000mL three-mouth bottle, stir, drop 160.8mL of perchloric acid, react at 10℃ for 48 hours, cool to 0℃, add 1000mL of water, extract take (the remaining tert-butyl acetate is extracted as solvent), neutralize with Na2CO3 to pH = 8~9, separate the liquid, then wash with 400 mL1% Na2CO3 aqueous solution three times, and concentrate the tert-butyl acetate layer under reduced pressure to obtain 42g of oil Glu(OtBu)2 with a yield of 32.4%. Add 42g of Glu(OtBu)2 into a 1000mL three-mouth bottle, then add 400mL of water and 11g of CuCl2, raise the temperature to 50℃, react for 12 hours, and prepare Cu[Glu(OtBu)]x(x = 1~2); When it is reduced to room temperature, add 19g of tetramethylethylenediamine and 100mL of dioxane, and then adjust the pH to 8~9 with triethylamine, L-Glutamic Acid 5-tert-Butyl Ester was prepared.
